It's an odd one, as it was a manager and a part-time wrestler/part-time manager but: Sir Oliver Humperdink and Eddie Gilbert.
Back story: Gilbert managed the Nightmare in Mid-South. Humperdink arrived and Nightmare turned on Gilbert to join Hump. Gilbert soon left Mid-South for a little while and when he came back, he joined Hump's stable, content to be part of the "winning team."
Cut to ONE YEAR after Hump took over Gilbert's charges. Humperdink introduces his newest wrestler, Taras Bulba, who proceeds to demolish Perry "Action" Jackson. As Hump, Gilbert and Taras celebrate in the ring, Taras turns and beats the holy hell out of Humperdink. All the while Gilbert is screaming "One year! I waited one year for this!"

Heenan reading Andre the riot act at Wrestlemania 6 after losing the tag titles to Demolition. Heenan always had that quality that he thought he was smarter than the men he managed at all times so when the proverbial poo hit the fan it all came out in a violent rage. Again, Heenan loses his cool and gives Andre both barrels, forgetting that the man he's dressing down is over seven feet tall and 500lbs. Ultimatley, Heenan crosses the line and Andre slaps the living snot out of him. Another quality Heenan had, he made sure that when he got his he did something to deserve it.

Bearer turning on the Taker will always stick out as memorable in my mind. The look on his face before doing it was pretty chilling at the time I saw it when I was 10

I want to say Savage-Sherri because Sherri's outfit that night was for me, personally, one of the most memorable things ever in wrestling.
But I think that may actually be the right answer. Between the Savage retirement implications and Elizabeth's dramatic return, plus the simple fact that Sherri was smacking the crap out of a defenseless former WWF champion, it was just a huge, huge deal.
Savage and Elizabeth's split was less definite (remember, she was in a "neutral corner" at WM5 despite him throwing her across the first-aid room on the Main Event), so I have trouble counting that one. I liked all of the Heenan incidents, but they happened often enough that it wasn't terribly shocking when a Family member finally fed the Brain some knuckles.

It wasn't really wrestler/manager at the time, but I remember marking out when Mr. Perfect dumped the water on Heenan and agreed to be Savage's partner at Survivor Series.

Yeah, along with slapping them, Heenan had a bad habit of saying the wrong things as well. Selling off Hercules to DiBiase. Telling Orndorff to vouch for Rude as the best bod. For a Brain, he didn't really think things through all the time

I always liked it how he acted surprised when one of his guys turned. Like he didn't see it coming.

Lou Albano turning face against Moolah basically put the WWF on the map. However, as far as iconic images, I go with Andre turning on Heenan and riding the ring cart back to the locker room alone.

One that hasn't been mentioned yet is Jimmy Hart protecting the face of Brutus Beefcake in 1993, when his clients Money Inc were going to smash his face with the briefcase. Jim Cornette "turning" on the Dynamic Dudes was my favorite, if only because the Dudes were gullible to believe that Jim would actually stay with them instead of the Midnight Express. One of my favorites. Gordon Solie and Jim Ross had to shout how evil and disgusting it was, over the crowd that was cheering for it. My other favorite was Paul E. firing Madusa at Halloween Havoc 92, where he called her a hooker and a bitch, then jumped up and down declaring she was "FIRED!! FIRED!!!!!!!" and shoved her, which finally caused her to snap and beat his ass up. Sadly, that meant the end of the Dangerous Alliance, grrrrrr.

Lou Albano turning on Jimmy Snuka in 1982 when Ray Stevens delivered 2 piledrivers to Snuka on the outside. This catapulted Snuka into the #1 babyface in the WWF until the arrival of Hulk Hogan in 1984.
