harrison ford & carrie fisher in original star wars trilogy (as well as anthony daniels & kenny baker ;D)
dan radcliffe, rupert grint, & emma watson as the harry potter trio
elijah wood & sean astin in lord of the rings
and someone mentioned bill cosby and his "wife" on cosby show, I would also say cosby & malcolm jamal warner as they made a nice father-son duo

Vivian Vance and Bill Frawley in I Love Lucy.
Some great chemistry between the two if you ask me; despite the fact their real-life hatred for each other is well-documented. Hell, Vivian actually celebrated when she found out Bill died.

What two actors that have played a couple in a television series do you think had the most chemistry? Is no one reading this part? Anyway, my vote goes to Ed O'Neill & Katey Sagal on Married... With Children, and Tim Allen & Patricia Richardson on Home Improvement.

Bruce Campbell and Toby Maguire actually. All of their brief interactions with eachother are golden and makes me want to see them in a buddy comedy. Toby Maguire also had good chemistry with James Franco. In fact, Maguire had better chemistry with those two guys than he did with Kirsten Dunst. ;D Also, James Franco had great chemistry with Seth Rogen in "Pineapple Express."
