Off the top of my head I'll say Mr. Peabody And Sherman and Click. And the scene in Eddie Murphy's Nutty Professor when Sherman is embarrassed at Dave Chappelle's jokes.

So do these have to be good?
Because how about when Hancock suddenly stopped being a comedy and turned into a horrible movie?

Click, Big Daddy, and Marley and Me.
I remember when Marley and Me came out during my first year at working at a movie theatre. Two co-workers and myself were standing near the entrance way waiting for the film to end, it was a sold out showing, and we were waiting so we could get in there and clean it before the next showing. As we're standing outside I notice so many people coming out crying, depressed, and in my mind I am thinking "Well, were they laughing so hard that they were crying?"
Then I overheard a young lady say "The dog died" and I knew right then and there I had no intention of seeing the film.

Prince Avalanche. I watched this on netflix a few months back and it was really funny and strange, but also had some really touching, sombre moments.

Barnyard
Mom and I took our cousin to see this. A PG cartoon-comedy starring Kevin James as a cow, and Sam Elliot as his father. 25 or so minutes into this generally silly coming of age tale, the latter dies of mortal wounds.

A Series of Unfortunate Events
Most of the film is black comedy, but the bit at the end where the Baudilaires find their letter from their dead parents is genuinely sad.

That was supposed to be a comedy? It felt like it... that was just a weird movie. Here we go on a long ride, where the movie jumps all over the place and tries to be a drama, but doesn't really hit it until after a hour in. Just an odd movie.
