Duhamel keeps losing parts because every casting director thinks they're getting Timothy Olyphant. Then they're like, wait, you're not Raylan Givens.... This was my joke! He looks too much like Olyphant and doesn't have a fraction of the talent. There is your issue. Alsl, how did JGL get mentioned? Dude is a legit A Lister that has worked with Nolan, Spielberg, Zemeckis and Stone, while starring in comedies, dramas and action movies. What the hell else is he supposed to do?

Billy Zane, the guy was amazing in Titanic, looked like he would become a huge star after it but cant recall ever seeing anything else with him since.

Thora Birch, a wonderful performance in American Beauty should have set her on the path to stardom, but spoiled by her overly controlling father practically blacklisting her from Hollywood.
Similarly Kate Hudson launched onto the scene with Almost Famous, and then through laziness has become stigmatized as a bad rom-com star when she could and should be far more.

I was a kid when this show was on, but I thought Jenny Garth and Ian Ziering would go on to be big stars for years after Beverly Hill 90210, especially Jenny Garth. Also, I thought Bridget Fonda would become more successful and famous than she ended being. I thought Sam Neill would get a much bigger boost following Jurassic Park.

Sam Worthington is going to be in Avatar 2 and 3 so his career hasn't hit the dumps yet. But it's also because he's not that good of an actor and can't emote well at least from what I could tell.

David Allan Grier. He was terrific on On Living Color and it just seemed a foregone conclusion he would break out. He just never did. He was also pretty damn terrifying in Tales From The Hood too.

Another one who just came to mind is Jason Sudeikis.
In 2012, on this very forum, I said Sudeikis had "Will Ferrell potential" and "all the makings of a star." And while he's put together a nice career outside of "Saturday Night Live", with starring roles in "Hall Pass", "We're the Millers", and both "Horrible Bosses" movies, he's nowhere near the level I expected him to be. I thought for sure by now he'd be able to carry a movie to a $75 million box office on his presence in a movie alone.
Granted, he's only been off SNL for a few years now, but it took Ferrell about two or three years before he was starring in "Old School", "Elf", and "Anchorman". From a cursory look at Sudeikis' filmography, it doesn't appear that he has anything resembling those sorts of roles on the horizon.

When Full House first came on the air Dave Coulier was looked at as the breakout star of the show. Unfortunately his act quickly became pretty obnoxious. Yes he still gets regular work, especially his VA stuff, but he never became the huge star I think a lot of people thought he'd become in those early days.
Speaking of breakout stars, Dana Carvey was considered the breakout star of the "Wayne's World" movies much more then Mike Myers was. But his choices for follow-up projects were pretty much disastrous on every level. He shows up once in a blue moon just to remind us he's still alive, but I don't think he'll ever have any kind of significant part in any project ever again.
After "Eight Mile" became a hit everybody started talking about how Eminem could be this great crossover actor with a big movie career ahead of him. This was luckily quickly forgotten about.
On a more personal note, I really thought after Smallville that Allison Mack would be one of the biggest movie and TV stars in the world today. Most of her work though nowadays seems to be obscure art house stuff, although seemingly she's done this by choice.

We're the Millers out-grossed all three of those Ferrell films though, so I'd say he's doing pretty good, even if he wasn't top-billed in that film. I may be wrong but I don't recall a single film in which Ferrell has been the main star out-grossing We're the Millers (with the possible exception of The Lego Movie, if that counts).
