I'll court hate and say, Ted McGinley on Married with Children. I wasn't a fan from the beginning, so I don't have a ton of Steve love, but for me, watching episodes back, they really started to hit a groove when Jefferson was added. Sure, it became more cartoony, but once it made that switch, it added more seasons to it's run.

On a similar Star Trek note, Jeri Ryan for Voyager. Certainly prolonged the show at the very least, I don't feel they'd have gone so long with the level of dwindling interest and story material they'd had for the previous three and a bit seasons or so.

Breaking Bad wouldn't have been as good as it was without Bob Odenkirk as Saul Goodman. It was in its sophomore year and hadn't taken off among the mainstream yet when they added him.

Supposedly Reginald Johnson and company were upset at first that their sitcom was overtaken by what was suppose to be a bit character. I'm sure they didn't mind the ratings and money.

I'd say you're partially correct, as her chemistry with Robert Picardo was really the beating heart of the show for the second half of its run.
