I assume this doesn't count the Adult Swim originals?
If not, then Dexter's Lab, Powerpuff Girls, and hell, Adventure Time and Regular Show are up there, too. I also liked Ben 10 a lot, though I haven't really followed the current spinoff.
Also while not 100% original, Justice League and JLU are original run on Cartoon Network (and CN's owned by DC's parent company anyway). And both are excellent.

Adult Swim: Either Harvey Birdman or Aqua Teen Hunger Force. Both are great.

Samurai Jack, Dexter's Lab, Flapjack, or Regular Show.
All four of those are very different in style, but they are basically the pinnacle of what you can do in animation for different reasons.

Either Dexter's Laboratory or Regular Show - slight edge to Dexter's Lab because it still holds up today IMO.

Even if you add the Adult Swim shows, I don't think any of them are even in the top 5, let alone #1.
I ALMOST wanna put Megas XLR in there somewhere, but the quality of season 2 was just off. It was insanely good at the start, but it was like they only had a few good ideas for the show and never thought past that.
#3 would be Johnny Bravo. #2 Regular Show and #1 is Dexter's Lab.
As for the action DC-ish shows...I'll fess up, I liked Teen Titans more than any of them.

Pretty much any of the Original CARTOON CARTOONS should be on the list somewhere (Dexter, Johnny Bravo, PPG, Cow and Chicken, Eds, Courage)

If we're counting Adult Swim originals then my choice would "The Venture Bros" nothing even comes close. I mean they (Jackson Publick and Doc Hammer) took what could have been a one note Johnny Quest parody and turned it into a hilarious show with a deep ever expanding world and mythology.

Samurai Jack or Dexter's Lab, they're both so good I can't really choose.
Johnny Bravo, Cow & Chicken, Ed Edd n Eddy, Courage, Billy & Mandy, the original Ben 10 (the following two shows are terrible), Adventure Time, PPG, Foster's, and Space Ghost Coast to Coast (this technically counts, right?) are in the running. I like what I've seen of Regular Show but haven't seen enough of it.
