If you can truly call JJJ a villain JK Simmons is the embodiment of perfect casting.

A cheat, but Kevin Clash to me is the iconic voice of Splinter. Not bad for guy best known for being the voice and puppeteer of Elmo.
Kelsey Grammar as Beast was also so good, he manages to more or less carry a shitty X3 film.

For heroes, its a close call between Christopher Reeve as Supes and Robert Downey Junior as Iron Man. I think the latter's portrayal of the character went a long way towards rehabilitating him post-Civil War.
For villains, Burgess Meredith as the Penguin.

Agreed on RDJ as Tony Stark and JK Simmons as Jameson. Chris Evans has been an amazing Captain America, and was pretty good as Human Torch (which ain't bad, considering how different the characters are). Loved Alfred Molina as Doc Ock.
Also, not necessarily a superhero, but Michael Gough will always be Alfred to me.

Chris Evans as both Captain America and Human Torch. The only highlight in terrible FF movies, his Johnny Storm is the quintessential portrayal of the character for me.

I won't say they are the best casting choice ever in a super hero movie but those two don't get enough praise in these kinds of threads. But JK Simmons as JJJ is the greatest character casting in any super hero/comic book/graphic novel movie ever. He better reprise the role in the new Spider-Man films

Andrew Garfield as Spider-Man. First one to get that balance of wit and angst right, and is almost believable as a character who's meant to be the put-upon nerd but is nonetheless able to marry a literal supermodel.

There has never been a better cast superhero than Christopher Reeve as Superman, nor do I think there every will be. He embodied that character and truly made you believe he WAS Superman. The way he carried himself, his manner of speak, the kindness in his eyes. He was Superman. He also somehow managed to make a pair of glasses and a slicked down hairstyle a believable disguise by making Clark a completely different character.
As villains go, I'm going with the OP and counting J. Jonah Jameson as a villain. J.K. Simmons brought that character to life in such a way it was almost scary. As if someone used some kind of magic to rip him from the pages of Spider-Man and turn him into a living breathing person.
Now, as straight-up villains go, no one can touch Jack Nicholson as The Joker as far as I'm concerned. A lot of his detractors will claim it was just "Jack playing Jack", which is absolutely false if you've ever seen Jack Nicholson as himself. The man is very calm, very serious, and very still. His Joker was manic, and full of glee with everything he did. Ledger was a great movie villain, but he didn't embody that joy of evil the way Nicholson did.

Samuel L. Jackson as Nick Fury Does this even count? The modern character is based on Samuel L, so it's basically just Samuel L playing himself.
