Super Best Friendcast is a fun one. Mostly video game focused, but with some hilarious digressions, such as Pat declaring Ariel the worst Disney princess for just "Wanting the D", and his horror a few weeks later when his dad brought it up to chastise him as Pat's sister has Ariel as a favorite. He had not anticipated his parents would listen to the podcast at all.

A lot of my favourites have already been mentioned.
We Hate Movies is phenomenal. It helps when you've seen the movie in question, but it's certainly not a prerequisite. If I were to pick an episode for a first-time listener, I'd go with Mortal Kombat or Mrs. Doubtfire.
I've been listening to Missing Richard Simmons recently. It's a pretty fascinating look at his exit from public life and if there's something nefarious going on that we don't know about. It's hosted by a former Daily Show producer who was a student of Simmons'.
