Now, I know some people hate the milking of sequels... but I can think of one that is begging for it right now: Bioware is basically THE in demand company right now, w/ Mass Effect being hyped huge, as well as the upcoming MMO Star Wars: The Old Republic. I've felt that Jade Empire is a very under-rated game, and their 1st original IP after doing KotOR. It may not be the BEST Bioware game, but a sequel on the Xbox 360 and PS3 could take the good things the game did and improve on them. What are some you wish would get picked up again? Agreed with everything posted so far.
Personally, I seeeeriously think the Zone of the Enders series needs another entry. It's one of my all-time favorite game series. I think the stories are awesome, the music is great, and most importantly, the gameplay is INSANELY fun. It's easily one of the greatest action series ever released, at least in my eyes.
The Fist of Mars was pretty cool too, but I suck at SRPGs, so I never got particularly far in it.

The World Ends With You – They’ve created an interesting world and only shown us a tiny part of it so it’s begging for a sequel that expands on the original and fixes its few flaws.
Little King’s Story, it was so close to being a classic I want it to get a second chance.

Mirrors Edge 2: I loved the first one and there's so much they can add to a sequel. Like giving it a bit of a sandbox style so you've got the entire city to travel across the rooftops with. Valkyria Chronicles: I know there are sequels for the PSP but I haven't got a PSP. I want a PS3 sequel!!! Knights of the Old Republic: An actual sequel not a MMO game that's limited to PC only so I'll never play it. X-Men Legends: Would love an X-Men legends 3. There's a whole range of X-Men storylines they can use or tie into it. Like the Morlock Massacre which would make for a good few levels and the Dark Phoenix Saga. Marvel Ultimate Alliance: ^^^ Likewise Red Dead Redemption: A sequel wouldn't work imo since RDR was at the final stages of the old west. A prequel though would be good fun. Nuclear Strike: I loved the Strike games. On Nuclear Strike they teased a sequel called 'Future Strike' but nothing ever came of it
