At least of the non-monster/superheavyweight (e.g. Yoko, Mankind) variety? Punk has been really sort of meh in terms of muscle definition for the last few years, which Triple H so helpfully pointed out, and I think even the Miz and Jeff Hardy had him beat on pure definition during their runs.
Not saying it's necessarily a bad thing, as the different MMA weight classes, for example, are showing that not everyone needs to be built like Brock Lesnar to be a believable fighter, just thought it was interesting especially if we're assuming Vince still has his bodybuilder fetish.

He's very muscular. However, he opts to do cardio in addition to his strength training rather than working on definition specifically. Kinda the opposite of Randy Orton.

I don't think Jeff Hardy had any muscle definition to speak of. Bob Backlund never had a six pack or big arms either.

Savage was really small when he won it the first time. Maybe even moreso compared to the people working at the time. I mean John Cena is basically a blown-up cruiser so it doesn't look ridiculous.
Shawn Michaels was also really small a few times. And no one is going to confuse Bob Backlund with Scott Steiner although he was in great shape.

I'd say Rey or JBL. Punk is like Edge body-wise, IMO. They have muscle and it shows but compared to the people they are facing it isn't impressive.

I'd argue Miz having more definition than Punk during his reign. These days? Maybe. Back then? He still had the flabby belly he's had for most of his career. Not that I particularly cared, just saying.
And, yeah, what others said, Backlund, Hardy, there's been others.

Jeff Hardy and HBK were wiry, yes, but they did have a fair bit of definition. Not counting the superweights, Miz probably fits the bill best, in part because he's basically a cruiserweight himself.

Punk's got a long body but he's fairly thick, and without the tattoos, one would notice that he's probably about as cut as Sheamus.
Jeff Hardy, HBK during his first reign and Bryan were far weedier. And as much as I love JBL, he was rolling in flab.

Sheamus's pale skin hides muscle definition far more than Punk's tattoos and yet Sheamus still looks more cut. Look at Pale Taker and Tanned Taker. It's for that reason that bodybuilders were bronzing lotion. Honestly I'd say Hardy is the only answer as far as during the time they were champion. Sure HBK, JBL, Flair and Eddie were all small (or flabby) at points in their careers but never as champion. '94 Backlund is the only other option.

Savage was really small when he won it the first time. Maybe even moreso compared to the people working at the time. He was small, especially compared to the giants working in the '80s, but he was still very muscular. He shrunk significanly in the early-'90s however.

What this OP should say is "Is Punk the least-defined WWE Champion?" because Punk definitely has muscle mass and general bulk, but he does have a layer of bodyfat or whatever that makes it way less prominent.

What this OP should say is "Is Punk the least-defined WWE Champion?" because Punk definitely has muscle mass and general bulk, but he does have a layer of bodyfat or whatever that makes it way less prominent. Yeah, I realized after I posted that I probably should've made it a little more specific. Because the superheavyweights like Foley and Yoko definitely had muscle under there too, but I tried to exclude them on general principle anyway since it's not even remotely a fair comparison.

I wouldn't say Miz is the least muscular. Miz may have been a bit flabby in the past, but he has always been pretty bulky. Miz has a wide frame and rather large pecs for someone of his stature.
