Austin running in for Foley's first title win immediately comes to mind. The crowd went nuts!
Also, Seth Rollins at WM 31, if you count a cash in as a run in, deserves a mention.

The end of this is just Bryan wrecking The Shield's shit on his own.

Kane returning to save Taker and Rock on RAW in 2000, after he'd been taken out by DX the previous week.

That time the Raw locker room ran in on Nexus leading to Nexus running away with Mark Henry somehow overtaking Heath Slater.

Lots of the best ones have been mentioned, but what about Kane returning to stop the UnAmericans from burning the US Flag?

If we are talking pure crowd reaction then it is Undertaker's run in on the bike at Judgment Day 2000 during Rock/HHH Iron Man. I was there and the place simply came unglued. I'm talking so loud you couldn't hear the person next to you. As far as the actual execution of it goes, well, I felt it was botched since the time was clearly up when HBK called for the DQ.
Austin's run in on Rock/Mankind is probably the best executed.

Brock Lesnar showing up during a Hardcore title match and just beating the shit out of Al Snow, Spike Dudley, and I wanna say Maven? has got to be up there, in part because he didn't even deign to pin them, He obviously came for a grander prize.

Kane's debut at the first Hell in a Cell match between HBK and Undertaker has to be up there.
As already mentioned, Austin's run ins for Mankind's first title win on Raw and at Backlash 2000 for Rock v Triple H would be my favourites though. Would give the nod to Backlash 2000 if I had to pick just one.

Because I see it quite often on here, would Arn and his Spinebuster from WM 18 count as a run in or interference?

It's got to be Austin saving the WWF from the Alliance in the build up to the Invasion ppv. That moment was just perfect with the whole show revolving around Austin's self doubt and then his big return, punctuated with him coming to the ring and just cleaning house with stunner after stunner. Goosebumps just thinking about it.
