Sgt. Slaughter went from defending his WWF Title against Hulk Hogan in the main event to being a member of an 8 Man Tag Team Match where I'm not sure he was legally tagged in for more than a minute. That tag match was a real shame. Neither team even got a theme music entrance on the PPV. Bossman and Duggan were over like crazy the last year or two and Nasty Boys were winning the tag belts the previous year. I do often wonder whose theme both teams came out to at the show. Yeah Bossman, Virgil, and the Nasty Boys all are candidates for this thread too. All collateral damage from the switch from four hours to three hours that year.

Hogan went from headlining Wrestlemania III to losing in a double DQ in his first match in a tournament the next year.
The Rock went from headlining Wrestlemania 17 to having to carry a returning 48-year-old.
You could really paint anyone who wasn't in the main event two consecutive years like this if you just go based off of looking at the card/results and not taking storylines into context.

Nikolai Volkoff went from winning the tag title to jobbing to Cpl. Kirchner in a incredibly short match.

Macho? Went from champ in main event to a mixed tag match, which was part comedy match? It was a prime match, being with Dusty and being King and a feature match being a first ever, but it’s still a big drop. Certainly not the biggest, but in the conversation somewhere. I’d argue it wasn’t that big a drop as the mixed tag was the second-biggest match on the card. With the exception of maybe Jake/DiBiase (and obviously Hogan/Warrior) it was the most hyped match of the night. The way Wrestlemanias were booked back then, the second biggest match on the card was in the slot just before the interval and the mixed tag was in that slot. This was also the case at WM6, 7, 8 and 9. My answer would be IRS. Went from being in one half of the double main event at WM9 to being part of a meaningless multi-man tag at WMX. And to top it all, the match got scrapped at the last minute and shunted to Raw the next night.

While not have an actual match, Papa Shango was involved in the finish of the main event at WM 8. For WM 9, Shango had a dark match with Tito Santana.

Jack Swagger - He went from having a world title match with Del Rio at 29 to working a tag match on the kickoff at 30. To make matters worse, he didn't even make the battle royal on the main show. You can also bring up winning MITB at Wrestlemania 26 and winning the World Heavyweight Championship two nights later. A year later he's seconding Michael Cole in the worst Wrestlemania match ever.

I argue a bigger plummet for Macho Man was winning the World title at Wrestlemania 8 to not even be on the card as a commentator for Wrestlemania 9.

Demolition Wrestlemani 6 , winning titles for 3rd time, getting some of the loudest cheers, to Wrestlemania 7 where they got no reaction (different music) and jobbed out in 5 minutes to a non full time WWF team

WrestleMania 25: WWE Title Match: Triple H (c) VS Randy Orton: BOTH guys plummeted and pretty hard. Triple H goes from defending the company's top title to fighting Sheamus in his first non-World Title match at WrestleMania since X-Seven. And, Orton goes from main eventing for the WWE Title to taking on his Legacy lackies.

If we're reading the question as "biggest loss of push/relevance with little hope of recovery," which appears to be the OP's intent, then Bundy (WM2 to WM3) and Demolition (WM6 to WM7) take this for me easily.
Honky Tonk Man from WM4 to WM5 is another candidate. He went from the IC champ no one could manage to dethrone to taking the pin in a filler match tagging with Valentine.

Demolition for three years straight was either competing or defending the tag titles, then in their last ever appearance they lose in three minutes to guys from a different promotion in a filler bout. Hard to fall harder than that

Hogan, HBK, Bret, Taker, Money Inc and Skinner if you count him as a Doink…a few more than I thought when I hit reply… I was gonna say at least he was on the show…but now I’m arguing Taker, from wrestling Jake to wrestling Giant Gonzales.
