I can probably name a lot of these from Rescue Rangers, which may be as infamous as Saved By The Bell for the "we never see X again..." trope.
Oh! On Gummi Bears, Flint Shrubwood! Though he didn't look like he fit in on their show.

The trajectory of LOST is notorious: seasons one, four, and five were great, two was faltering, three and seven were pits. I just remember watching some of that awesome stuff in 4 and 5 and going, "Christ, just imagine how great this would be if Mr. Eko was still alive." Ana Lucia was starting to grow on me, and I wanted to see how Hurley/Libby would have gone, but...you know... Not short-lived (you saw her most of S1 and 2, then she appeared sporadically on other seasons), but Ruby from Once Upon A Time became severely underused. Yeah, I know, Ory was doing other things, but yeah! As for G1 Transformers....another technicality, but...face/tweener Outcast Blitzwing.

The abbreviated season 4 didn't help, and I think it's the reason Charlotte was kinda nothing and Naomi's story just ends... they still were loading their plate with more than they could handle. (I absolutely can't believe Zoe Bell just for THAT.) But hot damn it really is just so apparent though that they had real plans, now. Juliet's arc is maybe the best of any character on the show, and Faraday's arc is right up there, too. Miles and Frank became real characters who fels solid and 3d. I think it is not a coincidence this started about when Brian K Vaughan came on board, by the way. And compare all that to how dirty the tailies were done. They cleeaaaarrrly had plans for Mr. Eko (I think the "Others speak Latin" thing was going to tie into him... he becomes a real priest and then is the only one who can communicate with Jacob, or something), but they were just too nebulous and the actor walked, and I don't blame him. But man we coulda had Mr. Eko in good LOST that would been so great.

I wouldn't have minded seeing Shelby and Jellicho show up in more episodes of Star Trek. Jellicho verbally sparring with Sisko would have been a delight. Oooh, Jellico during the Maquis or Dominion War storyline would have been interesting.

I alluded to this in the Paul Sorvino thread but I would have liked to see his character or George Dzundza's go for a longer time on Law & Order. I know they both chose to leave but I feel like the acting really took a nose dive since Jerry Orbach relied heavily on humor for his character rather than just playing a straight laced, great cop. I know Dick Wolf had to change things or be forced to cancel in 1993 with letting two actors go so there was probably pressure to change the direction of the show and it probably may not have been able to sustain 20 years with Paul or George but I'd have liked to see most of those 20 years with one of those guys in the lead role, especially Paul.

Nightbird from Transfomers G1. Yes, I'm sure she's made appearances is some comics or something elsewhere, but it would have been interesting to see her be a recurring threat. Especially since she pretty much punked out the Autobots in her one appearance on the show. OG Transformers had a lot of interesting characters that only appeared once or twice, like Kremzeek, Devcon the bounty hunter, Tracks's breakdancing friends, Powerglide's rich girlfriend, the mermaid that Seaspray fell in love with, the big game hunter who wanted Optimus Prime's head, and Elita One's all-female Autobot crew. I would have liked to see Circuit Breaker make a transition from the comics to the cartoon, but I suppose her backstory is a bit dark for that series anyways (well, pre-movie anyways).
