If CM Punk never wrestles again, would he be the only wrestler to be world champion after his final match, be stripped of the world championship (or have to give it up for another reason) and not wrestle another match after that? Closest I can think of is Edge who retired as world champ in 2011, but he did come back 9 years later.

Dr. Death Steve Williams won the “Asylum Championship Wrestling” world title in his last match before retiring after the match (due to his cancer)

With the qualifier of having to give the belt up, I can’t think of anyone. As far just retiring as champion, there’s Verne Gagne, and years before that, Frank Gotch.

If we're counting women as well, Gail Kim retiring as Knockouts champion has to be on the list, though she did un-retire to put Tessa Blanchard over.

Orville Brown was voted the first World champion of the National Wrestling Alliance; had to vacate after his career ended on November 1, 1949, following an automobile accident. He forfeited the title to Lou Thesz, as they were slated to have a match November 25th.
Danny Hodge vacated the NWA World Junior Heavyweight title after breaking his neck in an automobile accident on March 5, 1976; 3 days after defeating Hiro Matsuda for the championship.
Leroy McGuirk, on February 7, 1950, was in an auto accident of his own, and vacated the World Junior Heavyweight Championship after he was rendered blind in both eyes.
(Edit note: Hodge did come back for 1 match in 1983, putting over SWCW US Junior Heavyweight Champion Eric Embry in a match.)

Hector Garza was the Mexican Heavyweight Champion at the time he retired due to his battle with lung cancer. At the time of his death a year later, he was still officially champion.

Atsushi Aoki (AJPW Junior Heavyweight Champion) was another who died (motorcycle accident) while holding the title, June 3rd, 2019. The company would not officially end the reign until November 20th.
