Ok, I was just thinking of this due to the Roseabbe Becky thread and figured this deserved a more wider discussion.
But as I mentioned before, I hadn't even noticed the difference in the two Beckys on Roseanne for the longest time.
Other times it went largely unnoticed were in Bewitched with Darren, and well that's the only one I can think of off the top of my head.
Times when I did notice were: (not counting Doctor Who series) Doctor Wier (Stargate SG1/Atlantis) Quinn Mallory (Sliders) George McFly (Back to the Future I & II) Marty's girlfriend Jennifer (Back to the Future I and II & III)
Times I swore they changed actors but didn't: Lincoln's son LJ (Prison Break) John Connor, because of the different hair (The Sarah Connor Chronicles)
Times I'm still unsure and would need to look up: Quinn's mom and dad (Sliders) Tina (Quantum Leap) Rosco P Coltraine (Dukes of Hazzard)
And just because I simply haven't seen it recent enough to recall the character's name... the mean cop from Police Academy who always ran around with a guy who I think he called Proctor for some reason. Lasard comes to mind, but I think that was the Commendant(sp?) guy who had the role seemingly tailor made for Leslie Neilson.
So feel free to share any actor/actress replacements that you recall or heard of. I'm curious which ones went more largely unnoticed, especially where if they were main stars is concerned.

The mean cop was not recast, but rather they alternated mean cops. Proctor still was the mean cop of the day's asskisser though.

A couple off the top of my head are the moms in both Family Matters and Fresh Prince.

I don't know if it counts since it's a cartoon, but Lacy Chabert was the voice of Meg Griffin on Family Guy during Season 1, then replaced by Mila Kunis for Season 2 onwards.

Templeton Peck was replaced after the A-Team pilot (two-parter) from Tim Dunigan to Dirk Benedict. It was an obvious change. Dunigan looked sorta like Vinnie Barbarino and way too young to have fought in 'Nam. Dunigan is most known for being Captain Power on Captain Power and the Soldiers of Fortune.

Red Dwarf has done it in more then one occasion.
First when the droid Kryton made his first appearance in series 2 he was played by a different actor in rather shabby costume to be honest. When Kryton became a main character in Series 3 he became more robot looking and was played by Robert Llewellyn
Next there is Kristine Kochanski in series 1,2 and a episode in 6 was played by Clare Grogan. When series 7 came along the character like Kryton became a main character and was replaced by Chloe Annette.
But probably the biggest cast change to a character was the ship's computer Holly. For the first two series he was played by comedic actor Norman Lovett. After series 2 wrapped up Lovett did not want to continue with the series so he was replaced by Hattie Hayridge who played Hilly from the series 2 episode 'Holly Hop Drive' (In that episode the crew end up in a alternate reality which is the same as ours but everyone's sex is the opposite. Hilly is the female version of Holly). Hayridge would play Holly for next 3 series. When the Holly character returned at the end of series 7 Holly was once again played by Norman Lovett.

Laurie in that 70's show. The replacement was just horrible, did not do justice to the original at all.
