novel characters on film?
Im thinking it's Chris Evans, but I could be wrong.
He was in Fantastic Four, The Losers, Scott Pilgrim and the upcoming Captain America.
4 seems like a lot.

If we can count TV projects as well, John Glover's in the running: The voice of The Riddler on Batman TAS, Lex Luthor's father on Smallville, and The Floronic Man in BATMAN & ROBIN.
Neil Patrick Harris has voiced The Flash (JUSTICE LEAGUE: THE NEW FRONTIER), Nightwing (BATMAN: UNDER THE RED HOOD) and Music Meister (Batman: Brave and the Bold).
As mentioned, Ryan Reynolds has a few: Hannibal King in BLADE: TRINITY, Hal Jordan in GREEN LANTERN, and Wade Wilson in X-MEN ORIGINS: WOLVERINE.
And his ex-wife Scarlett has a few as well: Silken Floss in THE SPIRIT, Black Widow in IRON MAN 2, and Rebecca in GHOST WORLD (based on an indie comic).

The Joker aside, Mark Hamill was even in a movie called Comic Book: The Movie. He also did an inspired performance of one of the best comic book villains ever: Cock-Knocker. ;P
