Look around what the other boroughs have to offer that you'd be interested in, there's tons of things in Brooklyn, Queens, and the Bronx that can be an entire trip worth their own...and as a resident of nearby Jersey City I'd say there's stuff to do on the Jersey side of the river, too.
But yeah, a lot depends on what kinds of food you'd like to try, what kinds of museums you're interested in, if there's any particular kinds of stores that strike your fancy, etc. Being around 42nd means you'll have plenty of transit options, though, so you're in good shape to get anywhere in the city and surrounding metro.

And you'll probably get as many answers as people answering as we all have our favorites lol. Ray's sort of has the reputation, but there's also like, a gazillion different places claiming to be the "real" Ray's and I lost track of which one is actually which.
Just don't do a chain and you should be alright. But Scarr's in the Lower East Side is fantastic if you don't mind a schlepp downtown from where you are.

Other fun places to go if you're an anime, TV, or video game nerd:
Kinokuniya - 6th Avenue between W 40th and W 41st Street, right across the street from Bryant Park. A great Japanese bookstore with a big selection of manga, Japanese video game, and anime merch.
Nintendo NY - W 48th Street between 6th and 5th Avenue in Rockefeller Center. A huge Nintendo store with lots of games and exclusive merch. Worth going to even if you're not going to buy anything just to see the stuff there, including an original Game Boy that was melted by mortar fire from the first Gulf War. And it still works!
Midtown Comics - W 40th and 7th Avenue, right off Times Square. Excellent selection and they get a lot of in-store signings from artists and writers.
Chinatown Fair - 8 Mott Street in Chinatown. This little hole in the wall arcade is one of the Meccas of fighting games and other competitive arcade games.
Chinatown in general is pretty neat to walk around in, though there's a lot of uh, bootleg stuff to watch out for, too, lol.
There's a great little collection of Japanese stores and restaurants in Industry City in Sunset Park in Brooklyn. A few blocks up from that is the Captain America statue, honoring Brooklyn's favorite son.
If it's warm enough for a day spent outside, the Bronx Zoo is amazing.
My favorite museums are The Metropolitan Museum Of Art (5th Avenue around E 82nd Street, just inside Central Park on the Eastern side), American Museum of Natural History + Hayden Planetarium (Central Park West between W 77th and 81st Street), and the Paley Center for Media (W 52nd Street between 6th and 5th Avenues).

it will cost money, but get to a Rangers game at MSG if you can. Cost money ain't the word MSG might as well slap you for how disrespectful their ticket prices are Concession prices, too. Blew my mind when I was there for UFC 205.

So I bought a ring. And booked a carriage ride through Central Park. We've been together almost 20 years. Gonna ask her to marry me....again.
Also, we're doing the wax museum, going to Joe Bastianich's restaurant, Statue of Liberty/Ellis Island, gonna see Jerry Seinfeld at the Beacon and the rest of the trip will be more touristy stuff.

Oh as far as best pizza goes… I don’t expect you to go to Queens for pizza, but Amore in Flushing (my hometown spot) is fantastic

Oh! And if you ever find yourself in the southern part of Brooklyn along the shoreline, Roll N Roaster in Sheepshead Bay has amazing roast beef sandwiches. Corner of East 29th and Emmons. Letterman retiring killed any idea I’d ever entertain of visiting, but had I, I’d 100% go to Hello Deli. The first time I ever went to New York, I went there and had a sandwich (it was good) and got a picture with Rupert.
