Expected this as soon as the news came out Expect the same for Solar Opposites. Not exactly sure where this puts High on Life, but if it gets a sequel or updates I assume he's recast High on Life is a bit...trickier. Because Roiland literally founded the game studio that made it. It's also self-published so it's not like a bigger publisher can put pressure on the developer to get rid of him.

Microsoft maybe could with how aggressively they've helped promote it, but really any attempts they could make to do so likely would have been more effective before that shitfest game was inexplicably a hit.

As has been mentioned, weird, respectable kudos for Adult Swim if they were the ones who made the call to force him out. I can't recall how much he was involved in the writing process, but again, as has also been mentioned, at least Harmon seems to have gotten his act together and is arguably the funnier one anyway.
Also it's not like the lore of the setting wouldn't make it easy enough to come up with a justification for the voices changing, if they even draw attention to it at all. R&M is meta enough and it's a big enough elephant in the room, that I totally see them doing it, though.

Is Rick and Morty the first mainstream cartoon to have to recast its most important characters? Closest situation I can think of is Ren and Stimpy but that at least kept Billy West through both the John K and Games Animation eras.

Kinda, but Lacey Chabert was only the voice for the first season; I'd argue way more people associate Mila Kunis's voice with Meg than anyone else. Plus AFAIK Peter and the rest all stayed the same. The Justin Roiland situation would be like if Trey Parker had to leave South Park.

If you want to count in terms of deaths, maybe the most notable first time was Fred Flintstone going from Alan Reed to Henry Corden after Reed's death.
