Bryan Cranston is a good example. Don't get me wrong, he was hilarious as Hal on "Malcolm in the Middle," but I think his role as Walter White on "Breaking Bad" really helped people forget about Hal. You mean Dr.Tim Whatley?

Gonna stretch the idea a bit...
Ron Glass. Regular and very funny cast member of the classic sitcom "Barney Miller", and cult stardom status as Shepherd Book in the sci-fi "Firefly".

John Corbett was a key part of the ensemble on Northern Exposure but is probably more well known today, at least in terms of works in television, for Sex & The City.

Some GoT ones: Before she was Cersei Lena Hedley was the lead in The Sarah Connor Chronicles. Sean Bean was in Sharpe playing the lead role in a drama about a soldier during the Napoleonic Wars (based on books written by Bernard Cornwell, who wrote the series of books the Last Kingdom TV show was based on). Davos and Sam recently stole the show in the impressive Three Body Problem on Netflix. Richard Madden (Robb Stark) has since had lead billing on The Bodyguard (good) and Citadel (terrible). As well as his much memed Bane cameo Littlefinger had quite a large role on Peaky Blinders and first came to my attention when he played an annoyingly up his own arse (and of course handsome and successful so probably the writer's self-insert fantasy) character in the original Queer as Folk.

Courtney Cox with Friends and Cougar Town
Terri Hatcher: Lois and Clark, Desperate Housewives
Laura Prepon: That 70s Show and Orange is the New Black
A few of these examples are more "shows they've been in"
I'm taking it more as "show they had a core role in"
