I'd have him lose to Dean Ambrose. The match would be an all-out fight with both guys pounding away on each other. Brock would keep taking Dean down, but no matter he would somehow survive and get right back up. He'd catch him off-guard a few times, just enough to where you think he might have a chance, but Brock too would shrug him off.
Brock would eventually hit him with the F-5, but Dean would get the shoulder up. Dean would nail him with Dirty Deeds, but Brock would get his foot on the ropes. Eventually we'd get to near the thirty minute mark with both guys running on fumes at this point. Ambrose would manage to counter another F-5 attempt and turn it into a rear-naked choke. Brock would desperately fight, trying to shake him off, even ramming him back against the turnbuckle to try and do so, but Ambrose would clinch it right back on every time, refusing to let go.
Eventually Brock would drop to the mat and after fighting with everything he's got, he'd pass out.
And Dean Ambrose would become the new WWE World Heavyweight Champion.

You could try and replicate the Extreme Rules match with Ambrose - Extreme Rules match where Ambrose is savvy with weapon use. He's a lunatic who makes no apologies for doing whatever is needed so weapon use is encouraged by his character and Brock has been shown to be weak to weapons as he lost the Extreme Rules matches to Cena and HHH.

The levels of carnage would be unreal. Hell, I'll say that with every active talent in the WWE today, a No Holds Barred Street Fight between Dean Ambrose and Brock Lesnar is my dream match.

Honestly, I'd say the best way for Brock to eventually lose the title would be an I Quit match.
He's the guy with the reputation of believing he's better than the business. He's a mercenary. He only cares about himself. Having that belt brings him the pride of knowing he's better than the others. He doesn't care about linage or legacy. He cares about showing "Look, I'm Brock Lesnar and no one can stop me."
Put him in a match where he straight up murders another human being, but they won't quit and eat more and more damage while getting in a few shots of their own. Finally they trap him in a predicament where he can't escape (maybe he charges the corner, misses, and they handcuff his hands allowing a Boston Crab like move with the top turnbuckle as added pressure) and he quits to save his self (Bonus points if Heyman is cut off by another person before he can use bolt cutters to free Brock's hands).
In the end, they couldn't out power him, but instead out planned him.

I predict he beats Cena at Night of champions,
Seth rollins music will hit,Paul Heyman will nod to brock as he lays down allowing Seth to easily cover him and win the belt.
Paul will leave with Brock and shake Triple H's hand as they exit, Meanwhile Triple H claps as he comes down to celebrate with Seth Rollins.

Just let the two monsters level Tokyo, but I want to see them in an last man stand match. That match would be awesome and as much as Lesnar is an asskicking machine I always bet on Haku/Meng

Wade Barrett challenges the Authority, wondering why he's been forgotten. Demands a title match. They say it's a death sentence. Over the next couple weeks unrest grows in the WWE locker room as Lesnar begins terrorizing the back, interrupting matches, saying he can do whatever he wants; he ruins big matches, like for instance he'd just have come out in the middle of Ambrose/Rollins and F5'd them both, like he violently buzzkills everything. PPV comes. Wade Barrett vs. Brock Lesnar, no holds barred. Brock scores early super bad beatdown, viciously destroying Barrett like he did Cena. German Snap German Powerbomb German Mounted Elbows German German - MEATHOOK CLOTHESLINE BROCK IS DOWN RYBACK IN RING - - SLATER! SWEETNESS AS BROCK TRIES TO STAND - - BROCK POPS UP - ELBOW FROM BARRETT- - 450 GABRIEL OFF THE TOP - 1 2 3 Barrett is champ. Triple H, Stephanie, Kane and Paul Heyman on the stage going insane. On the night of the match, HHH has Brie kidnapped and orders Daniel Bryan to lose the fight to save her life. As the battle begins, Brie manages to escape, but just as Hunter's goons corner her, a repentant Nikki rescues her by knocking them out with a fire extinguisher. Lesnar has the upper hand at the start of the fight, ruthlessly pummeling Daniel, even trying to kill him by ripping out one of the steel posts and trying to run Daniel through. Brie cheers to urge Daniel to fight back. Daniel is re-energized by her words and once he sees Brie is safe, gains a second wind and starts to turn the tides on Lesnar. The fight destroys the ring, with Daniel and Lesnar continuing the battle up the through the stands as a disbelieving HHH watches from the control room with Stephanie McMahon. Daniel finally puts an end to the match by knocking Lesnar off a catwalk, which causes him to fall through the ring and lose painfully, as this was a Last Man Standing Match. A frustrated Hunter begins destroying electrical equipment, in total disbelief over Lesnar's loss. Bryan angrily goes after Hunter intending to repay him for his misdeeds. In fear, Hunter retreats, but accidentally touches live wires he exposed in his tirade and is electrocuted to death; a distraught and emotionally wrecked Stephanie weeps on her knees at the sight of her fallen husband. With his enemies now vanquished, Daniel celebrates his victory with Brie and his friends.

Of the current roster, it has to be either Ambrose or Daniel Bryan in terms of match quality. I'm not feeling a Roman Reigns / Lesnar match at all to be honest. Agree with comments of Rock / Brock 2. That would be a really big deal, although the outcome will be somewaht obvious. The build and match itself would be ace though.

It doesn't matter who, it doesn't matter when. All that matters is someone challenges Brock to a No Holds Barred match. Brock and Heyman laugh at this then starting talking up Brock's UFC career calling him the greatest No Holds Barred fighter of all time. The challenger then points out that this isn't a UFC No Holds Barred match, this is a pro wrestling No Holds Barred match, then proceeds to use every single old-school dirty heel tactic imaginable to completely embarrass Lesnar - weapon shots, run-in's from your buddies, powder to the eyes, rags soaked in ether, even pull out the Mounties old shock stick. After Lesnar is thoroughly defeated the challenger then announces that while Brock might be the greatest fighter of all time, The Challenger is the greatest wrestler of all time, and everything he just did to Lesnar is what pro wrestling is truly all about.
Remember, you don't beat monsters with headlocks and takedowns, you beat monsters with pitchforks and torches.
