American Housewife's cancellation got me to thinking about this. How the last season saw two regulars pretty much leave, and one role was re-cast. It reminded me of another ABC example, Once Upon A Time, which also ended with many of the regulars leaving yet the show continuing on nonetheless.
I KNOW there has to be a lot of other good examples, let's think of some.

I don't watch The Walking Dead anymore, but when I see someone watching a new episode, it pretty much looks umrecognizable aside from Daryl and Carol still being around.

The final season of Laverne & Shirley is best remembered for having no Shirley with Cindy Williams leaving, but people often forget that Betty Garrett (who played Edna, Laverne's step-mother) also left that season, and Michael McKean (Lenny) was reduced to a recurring character. He was credited as a regular in the episodes he was in, but that was only about half of them.

Walking Dead has let go of a lot of characters over time. Maggie, Daryl, Carol, Ezekiel, Rosita, Eugene and Gabriel are the ones that are left over but you're missing the most important point which is someone in the Grimes family not counting Judith
By the time Smallville ended, you were left with Clark and Lois as the regulars. Chloe was there but not featured in every episode while Oliver has been a regular since like Season 8 but some of his stuff was tied to Chloe.

Two things: 1) Didn't Edna leave the season before? 2) Actually one Lenny episode, the Death Row one, had the David Lander-only version of the opening credits.

I’m fairly certain none of the original Coronation Street cast are left, although the show started in 1960.

Maybe not the same thing, but Archie Bunker's Place just had Archie; Edith, Meathead, Gloria, the Jefferson, all gone.

MASH had only Hawkeye and Hotlips left as main characters from the original, first episode cast (I want to say Father Mulcahey was introduced later but even if that's not the case, the original was a different actor and didn't have any lines the first time he appeared).
NCIS will be similar when it ends, with Gibbs and Ducky the only major roles left from the first episode.

Grey's Anatomy currently has like 3 people from the original cast.... only 1 of the original interns. It's still kicking along as it was renewed. I swear, they are going to try and kill/remove the entire original cast before ending the series.

Wikipedia says she was in season seven so that's what I went with. Another show with a lot of turnovers over its eleven seasons was M*A*S*H, although the cast was pretty stable for its final few years. The only regulars from start to finish were Alan Alda and Loretta Swift. Jamie Farr and William Christopher were also there from beginning to end, but they were not regulars in the first few seasons. And I think everybody pretty much agrees that trying to continue That 70's Show after Topher Grace left was just a bad idea.

I also like how Hawkeye's hometown of Crab Apple Cove, Maine came up a lot but really early on they hinted he was from Vermont.

Are we counting Saved By The Bell: The New Class as part of the original show?

The British version of Being Human lost all its original cast about halfway through the series. Ditto Misfits.

Shall we explore the saga of Spearchucker Jones, or is that best left unsaid? Spearchucker was awesome and actually important in the book and movie. Then it was like, we want someone named that for the show for like 3 episodes. The tv one was great too, and you bought that he was Hawkeye and Trapper's equal, living in the Swamp too. The justification for the nick name never, ever worked though.

Sliders. Only one of the characters made it from the start to the end of season five.

William Russell will return. He's just biding his time for when it will have the most impact.

I’m fairly certain none of the original Coronation Street cast are left, although the show started in 1960. William Roach (Ken Barlow) is still there
