Funniest in my recent watching history is the Black Books episode where they decide to start serving food in the store to compete with the big chain.
"Where's my tower of soup?!?!?!?" Followed by the look on Manny's face had me dying.

The One with the Embryos from Friends is TV at it's finest. "It's actually Miss Chanandler Bong." 'The One Where No-one's Ready' is even better. Nothing short of genius, and virtually in real-time. Could I be wearing any more clothes?

Have to give an honorable mention to the COMEDY BANG! BANG! season 2 episode 'Gillian Jacobs wears a red dress' .. The episode features forshadowing to events later in the episode, a vampire chef, and a mystery to the contents of a coffin* * {Spoiler}It was Onions

Last Exit to Springfield. Every single moment has a brilliant joke packed inside. One day I was sitting around thinking of some of my favorite Simpsons moments. I then realized that around 80% of them came from this single episode.

'The One Where No-one's Ready' is even better. Nothing short of genius, and virtually in real-time. "Could I BE wearing any more clothes?" I absolutely love both those episodes , however the one where everyone finds out is my favorite of all time m.youtube.com/watch?v=xMWiirWWxjU

The Simpsons would probably claim the top 10 of my list at least but I think "Homer Goes to College" might top that list.
For all other shows, I'd probably go with Newsradio's "Airport". Quite possibly the crowning moment of Phil Hartman's amazing career.

The Teacher's Strike episode of Simpsons ranks very highly for me. There's always a joke I had forgotten by the time I end up seeing it again that gets me all over again.

The Captain Tuttle episode from M*A*S*H has gotten around 15 or so views from me and not once have I been able to keep smiling ear to ear and boiling into laughter because of it.
I've probably laughed way more in other episodes of other TV shows, but it's a pretty perfect episode of sitcom television, and I just mentioned because I don't think it'll get any love otherwise.

How I Met Your Mother's "Slap Bet" (aka "Robin Sparkles") and "The Bro Mitzvah" (Barney's Bachelor party) are the first to come to mind when I think of that show. Both were utterly brilliant.
Looking at my HDD, The Colbert Report's "Francis Fukuyama" episode is a stand out. It's the Bin Laden death one. Colbert had heaps of fun with it.

The episode of Cheers where Cliff Clavin goes on Jeopardy. That episode is great but I think the episode with John Cleese as a marriage counselor is just as good.

Followed by the look on Manny's face had me dying. The episode with The Cleaner and Manny and Bernard brewing a wine that ends up killing the Pope would probably get my vote, but Black Books has some definite contenders. Manny: You are a filth *wizard*. Friend only to the pig and the rat. Ugh. Look? [he opens a pizza box] Bernard: Pizza. I was going to warm it and eat it later. *Everybody* does that. That's *normal*. You are just *looking* for things to complain about. Manny: And what are these? Bernard: ...wasps.

Speaking of John Cleese, Fawlty Tower's "The Germans" is still one of the funniest things I've ever watched.

Slap Bet from HIMYM is a solid episode that made me watch the series. Nice physical comedy, good build up, and an amazing reveal that would have been perfect if the commercial lead in hadn't revealed it.
