Has there ever been a show where they replaced one of, if not THE main character/actor?
I was thinking of Michael J Fox today for some reason and it reminded me of Spin city and how he was replaced by Charlie sheen. Charlie sheen was replaced by Ashton Kutcher on 2.5 men. Ashton Kutcher and Topher Grace was replaced by that one guy on that 70s show.
Before that, I clearly remember liking head of the class and found it weird they replaced the teacher with some Scottish guy.
The only time I saw it work was Cheers as I greatly preferred the Rebecca years over Diane’s also preferred Woody over coach

Mark Ruffalo as Bruce Banner/Hulk, coming from Edward Norton. I don't think anybody missed Norton after seeing Ruffalo in The Avengers.

Danny Masterson was basically the co-star of The Ranch and was replaced with Dax Shepard, but how successful that was we will never know unless Netflix decides to share.
Dominic West was at best a supporting character in season 4 of The Wire, and that’s widely regarded as the best of the series. It was always an ensemble cast, but the show hit its stride when McNulty was out of the spotlight.

Valerie Harper was booted off her own sitcom in season 2 and then the show was called the Hogan Family and ran for four more seasons.

Well the thing about Cheers was that during the Diane years, the show was very much a "will they/won't they" show for Sam and Diane. They had basically taken that about as far as it could go when Shelley Long left. With her gone the show became much more of an ensemble cast and really breathed new life into it. I really believe that's why it went eleven seasons instead of only seven or eight.
I don't know if it counts, but I really thought The Daily Show was dead in the water after Craig Kilborn left. I mean, why would anybody watch the show without "Five Questions"? Or "Pudding Time"? Man was I ever wrong about that.
One of the earliest major recastings I can think of is Make Room for Daddy/The Danny Thomas Show when Jean Hagen's Margaret, the shows original wife/mother, was replaced after three seasons by Marjorie Lord's Kathy. Show went another eight seasons afterwards and went from a solid time killer to one of the most popular shows on television at the time.

Old allegations against him were brought up again two seasons into the show and got some traction, so Netflix politely asked him to bail and he agreed.

Zachary Levi (who was originally supposed to be Fandral in Thor 1 but couldn't due to conflicts with filming Chuck) played Fandral in Thor 2 and 3 replacing Josh Dallas (who had to leave due to conflicts with filming Once Upon a Time) >_>

Hattie Hayridge was just as good a Holly as Norman Lovett and saw Red Dwarf through many series. Though they later reverted to Lovett's version, hers is by no means a downgrade and indeed has many of the character's most iconic moments and got the characterisation perfect.

Howard did ask for more money, but rather than negotiate with something both sides could live with, Ike "Scum of the Earth" Perlmutter decided to recast and reasoned nobody would care because "black people all look alike." Seriously.

When Marvel first got the movie rights for Iron Man back from, I think, Universal, Marvel had absolutely no background in making their own movies and weren't really attracting any A-level names to the project. Finally they came across Terrence Howard who was interested, and they gave him a big money deal to play Rhodey, probably more then he was actually worth, with the hopes that Howard's name attached to the project would bring in other top names. That didn't really work out. Eventually Jon Favreau and Robert Downey Jr. agreed to do the movie, completely independent of Howard, simply because they were fans of the character. They wound up getting paid much less than Howard, and even when other big names like Bridges and Paltrow jumped on, Howard was still easily the highest paid actor in the first movie. The movie was a success and Marvel made plans for a sequel, but insisted on cutting Terrance Howard's pay. Now to Howard, he had just starred in a wildly successful movie that just made a gajillion dollars, he was being asked to return for a sequel with his part being significantly expanded, and was told he needed to take a pay cut to do it. To him, and probably most actors with any name value, this would sound ridiculous, so he walked, and the part was recast with Don Cheadle. Also remember at the time Marvel Films was under the direct control of the notoriously stingy Marvel CEO Ike Perlmutter, so that probably added to the issue. If I remember correct they very nearly lost Samuel L. Jackson for similar reasons.
