Many of these show, while quality shows, got canceled because they did not get the ratings that were acceptable enough based on their budgets. Shows like Firefly, Terminator: The Sarah Conners Chronicles, Caprica, Fastlane, etc all had significant budgets that required significant ratings to offset the expenses related to them. Other shows like The Defenders would have survived had it been on a network other than CBS where the ratings that it pulled in were considered significant.
I will list a couple of shows that should not have been canceled when they were based on ratings:
1) Just the Ten of Us - Some of you might not remember much about this spin-off of Growing Pains, but it actually was a hit show. It still won its time slot in its final season and proved a strong lead in for 20/20. It was canceled for political purposes, specifically to make Miller/Boyett/Lorimar happy. They canceled the only TGIF program not produced by them (Just the Ten of Us) to open the slot for Going Places, a new sitcom that Miller/Boyett were pitching the network, just so that they could have an entire block produced by the same company. The fact that they canceled a hit in favor of a show that did not even last an entire season got some people fired.
2) Scrubs - I know some people hate the last season of the show and some even refuse to consider it the same show. Amongst the later, just about everyone involved in the production of the series except the studio itself. The ninth season was intended by the producers to be a spin-off series and was produced as one. The problem was that the studio that owned Scrubs, ABC Studios, wanted to be able to add the ninth and any subsequent seasons as part of the lucrative syndication deals that they were getting for Scrubs. As such, the show was continued as was, likely complete with the same cost per episode to the ABC Network (as charged by ABC Studios). Had it been a legitimate spin-off it likely would have had a much smaller rate to the network. I would assume that the lower cost would have made continuing the series more palatable to the network when they looked at the ratings (which were improving towards the end).

Well it wasn't really canceled but the Wire should got a proper full 5th season as well as a final sixth season as well.

The Tick (live action show) - FOX didn't even give it a chance. Firefly is an obvious one. I'll never understand why Disney pulled the plug on making new Power Ranger shows and decided to re-master the original series. Thank goodness Saban brought them back.

I'll forgive The Tick. Because it was cancelled, Patrick Warburton went on to voice Brock Sampson in The Venture Brothers. May not have happened otherwise. If we're including animated shows (have seen a couple mentioned), I can't overlook Sym Bionic Titan. That show was just awesome, and was cancelled because there weren't enough merchandising opportunities. Big FU to Cartoon Network on that one.

Journeyman as it probaby would have got a second series if it wasn't for the writer's strike, admitedly a weak start didn't help but it was really good by the end and I think it would have got even better. I'm going to add Tru Calling to this list, that show was baller. Another show with a weak start, it improved massively after her rival was introduced and s1 had a great finale. One person trying to save a life while the other attempts to make sure they die was a great concept. Yeah I just like time-travel, Firefly is the main one though.
