Which would it be?
I would have to choose the episode where JR was shot. Just great writing. It was masterfully done, leading up to the final scene perfectly, with a truly awesome, sinister performance by Larry Hagman.
It really did deserve all the attention it got.

There are many Twilight Zone episodes that come close, but the Scrubs episode "My Screw Up" with Brendan Fraser wins it for me.

I Love Lucy - Lucy Is Enceinte. Lucy tries to break the news of her being pregnant but is repeatedly thwarted by interruptions. Finally she is able to let him know during a show.

There's so many great ones, including the I Love Lucy one which I still love watching even though I wasn't born when it aired. Having to pick one, particularly one that hasn't been mentioned, I'm going to have to go with the MASH finale. The ending of that show was beautiful.
However, I'd have to give an honorable mention to My Lunch from Scrubs. The ending scene of it was among the best things I've ever seen on TV.

"Vincent And The Doctor" from Doctor Who.
It manages, in a little under 45 minutes, to help introduce a new generation of children to Van Gogh's work, discuss the realities of depression and how it's not easily fixed, show one of the most beautiful bits of SFX work ever (the 'Starry Night' sequence), develop the character of the companion after a big shift to the status quo in the previous episode, and still have time for a damn good historical sci-fi adventure while doing all of this. The acting was top class all round, particularly from Tony Curran. It was the highlight of the series, and it reduced both of my parents (and me) to tears.

All In The Family - The Stivics Go West.
Seeing the Stivics leaving and seeing Archie of all people crying after Mike said goodbye really got to me. I'd consider that the best episode of the entire series.

"In the Pale Moonlight" from ST:DS9. An incredible episode that takes Star Trek to its darkest and most intriguing place and raises some of the biggest questions in the history of the franchise.
