Robert Llewellyn replaced David Ross as Kryten in Red Dwarf, and the show was all the better for it. There's nothing wrong with Ross' performance, but Llewellyn just brings so many little things to the role, from mannerisms to facial expressions to tones of voice, and I honestly don't believe the character would have stuck around if he hadn't been so brilliant.
Also, Ruby Wax as Betty Monroe from Rocky Horror/Shock Treatment. Granted, we barely saw any of the previous Betty at the start of Rocky Horror, but either way, Wax is that character to me now.

American Gladiators: Larry Czonka over both Joe Theismann and Todd Christensen. This so much. What's funny is that Theismann was the original host. Mike Adamle was just his partner. Todd Christensen was an improvement over Joe but not by much. Adamle and Csonka were just magic together. And then Larry left, replaced by Lisa Malosky, and then the show went downhill. No offense to her or anything. I liked Lisa, but she's just not Larry Csonka.

MST3K: Murphy over Weinstein (though I'm personally a huge fan of) And by that same token: Frank Coniff (As T.V's Frank) replacing Weinstien's Dr. Ernheart. Again nothing really wrong with Josh, but Coniff really nailed the sidekick role and made it his own.

The GM C Type, I think it was called, over the original GM from the Gundam series. The original's design just does not age well, from the odd green-blue, the round head, and the diaper-looking crotch section. The redesign just looks much better and more like a functional war machine.

Hagar replacing Roth. God I hate Roth. Terrible singer, crappy lyrics, however a great frontman. BOOOOOOOOOOOOOOOO Van Halen's old stuff is classic, Van Hagar just sounds dated to me.

Mike Patton replacing Chuck Mosley for Faith No More. Second this motion. Not only did you have the phenomenal vocals of Patton coming in, but Chuck was a very monotone, sub-par vocalist to being with.
