Off the top of my head in terms of ones I've enjoyed...
1. NYC 2. Orlando 3. Boston (kind of bias though) 4. Los Angeles 5. Philadelphia

1. Chicago 2. Washington DC 3. Detroit (by default)
....
I don't really travel much, and when I do it isn't usually to cities.
I have also been to Toronto and Seoul, which obviously don't count. Probably would put both after DC.

1.La Crosse, WI (Crazy Beer, Great Food, Great People, Wacky Party atmosphere. I swear it has more bars than people. I know I'm stretching the definition of Major City) 2.Chicago (Great Food, the aquarium and Natural History Museum are the bees knees) 3.Washington DC (History, but shit traffic) 4. New Orleans (History, Food) 5. San Francisco (History/Food/Just a fun place to hang around and spend too much money)

1. New York 2. Philadelphia 3. Orlando (I guess) 4. Canada.
Yeah I've not done as much of the States as I really should.
if this were worldwide I'd be able to name you shitloads.
Was planning to go to California last year so Los Angeles and San Francisco would have been on the list had it gone through.
But honestly, a certain someone is putting me off going State side anytime soon.. I'll leave it at that.

no order:
1. Oakland (I live there, but it's on the come-up as a tourist destination for better/worse) 2. San Francisco 3. Miami (south beach is as nice as Burn Notice makes it seem) 4. New York (Lower Manhattan/Little Italy was my favorite spot) 5. I'll co-sign on Chicago too.

The only two US cities I've spent significant time in have been Buffalo and Dallas. Outside of that, just flight connections and a trip to Detroit that I was barely old enough to remember.

1. Memphis, I grew up spending weekends in Memphis since it was just an hour away from home. It might get a bad rap but it's always gonna be like home to me. #GoGrizz 2. St. Louis, kind of the same deal as Memphis, spent my fair share of weekend afternoons in old Busch Stadium while growing up. 3. Seattle, I love a town where I can see tons of weird shit and walk the entire time. My brother and I go every few years to visit old high school friends that live there. 4. San Diego, I dunno if it counts since we only went to the beach the scenery, the freedom, and being there with my future wife made it a nearly perfect place. 5. Las Vegas, didn't drink, walked the strip, saw a bunch of stuff, and hung out with the Ladyhawk, it was a great place to be.

New York Orlando Washington DC Baltimore Philadelphia
I've been to Miami and LA, too, but didn't really spend a lot of time either. Miami was more or less a day trip and LA was more in transition to Anaheim. I really don't think of Anaheim as a major city or I'd put it #3.

1.Boston 2.Chicago 3.Omaha 4. South Bend
Omaha was pretty good, middle of nowhere, got asked multiple times why the f*** I'd gone there.
I'd imagine Orlando will be up there soon.
I basically went to the last 2 visiting friends, hence them being a bit more random.

I haven't been to that many, but from where I've been:
Washington DC NYC Anchorage Austin San Antonio
Honorable mention to Houston. I've also been to Detroit, Kalamazoo and Hartford (CT) but they wouldn't make the list or probably don't count as major.
